A series of original puzzles have been developed by Reiner who is an “expert in algorithmically generated puzzles,” so expect your brain to be aching on more than one occasion!
This sounds like Professor Layton game (Layton no fushigi na machi)

Well there is Can't Stop (2 player only no AI though) and Puerto Rico for the DS and of course quite a few classic boardgames, etc. in Clubhouse Games.

Yeah, that's true. But of the 40-some games in Clubhouse, at least 30 are really crappy. There are only two or three that I really like, and the interface makes playing them very slow.
I haven't gotten into the homebrew scene (yet?), but games without AI don't excite me, nor does Puerto Rico.
Wow. That was negative. On the positive side, I'm really looking forward to Marvel Vs. and the new Final Fantasy Tactics for DS.

One of my favorite homebrew games is Set. You can play against an opponent with a local wireless connection.

Samurai not sure this would work or not when I play it on the DS mabiweb it's a ton of scrolling for sure
Modern Art PC version exists I could never beet the AI no matter how many times I tried, probably better to play in person.
Tichu 4p wi-fi this would force people to play a bomb of 6's when one wished for a 6 for instance. This would only work if it had wi-fi which is pretty hard to implement but not impossible though
San Juan I enjoy Jim's San Juan for Mac and with the dual screen I'm sure there is enough screen real estate for card info, etc.
Acquire this would work most likely I've seen various PC, Mac versions of this
Lord of the Rings - The Confrontation this would never happen, but this would probably be my first choice for a dream come true

St. Petersburg PC version exists and it'd probably work on the DS
Quoridor (I wish some took xoridor source code GPL and ported it to DS, not for the AI though as the ARM7 and ARM9 can't hack it when a dual core CPU can't hack it either)

But Microsoft currently isn't opposed to put less important brands on DS or let their teams develop for it as they don't have a competing portable. See Rare doing Diddy Kong Racing for Nintendo or the Age of Empires game on DS.
