I'm storing some Xbox games for a moving friend at the moment, and he encouraged me to take any of them for a spin. One is Fallout 3, which I've been interested in trying, but the high praise for 1 and 2 (especially 2) around these parts makes me want to try those as well. If I play 3 first, will that diminish anything in the first two?

It does have a few mentions, and some groups from the previous games.
But FallOut 3 isnt so much story related with the first 2 then FallOut New Vegas.
I would suggest you give it shoot. But the older games, although very good dont compare with this in terms of gameplay. FallOut 3 is the best of them all.

All Fallout games feature different main characters. So there is nothing to spoil. The only thing I would do is play 1 before you play 2 just because of the improvements to the system you would otherwise grow accustomated to and miss them then.
